民国时期，国内最大的兵工厂发生离奇命案，死者被“幽灵子弹”所杀——犯罪现场找不到弹头一时之间，疑云密布，人心惶惶，此案堪称民国第一奇案。此案交由警局内负责管理档案的政务警察老包调查（刘青云饰演），担任文职的老包既不会拳脚功夫，也不懂如何开枪，却思想缜密，对破疑难杂案有独特方式。一心为民除害的热血警探郭追（谢霆锋饰演）不容于警局贪污腐败氛围，联手老包调查此离奇命案。如何开枪不用子弹？如何让子弹在穿入肉体的时候消失？老包和郭追在枪和子弹上做了各种各样的研究，包括向猪身体发射盐子弹、冰子弹等等，却没有任何满意的结果。随着案情调查深入，两位警探抽丝剥茧式发现恐怖阴森的杀人密室和离奇的造弹装置，其诡异程度让经验丰富的老包也瞠目结舌。就在“幽灵子弹杀人”真相揭开之际，老包、郭追却接二连三遭人追杀，警察小五（井柏然饰演）在开枪掩护老包、郭追逃亡之时惨死。结案期限迫在眉睫，算命姑娘小云雀（杨幂饰演）不经意间透露出“幽灵子弹”惊天秘密，两人不知这个“惊天秘密”是圈套还是破案线索，却发誓要以命相搏！案情终于水落石出，郭追和老包却发现各自的命运被无情捉弄，两人只能活一个的选择摆在他们面前……
